In-depth explanation of Steroids

Steroids are lipids with a fused four-ring skeleton made of three cyclohexane rings and one cyclopentane ring, often called the steroid nucleus. Cholesterol is the parent steroid, and other members include bile acids, sex hormones such as testosterone and oestradiol, and corticosteroids. Small changes in the ring substituents, such as a single double bond or hydroxyl group, can produce large differences in biological activity. Steroids are made in the body from squalene through a series of cyclization and rearrangement steps.

Going deeper: Steroids

Key ideas: Based on the four-ring cyclopentanoperhydrophenanthrene skeleton, three six-membered rings (A, B, C) and one five-membered ring (D), 17 carbons in the core. Include cholesterol, bile acids, sex hormones (testosterone, estradiol), corticosteroids and vitamin D precursors. Trans ring fusions make them nearly flat and rigid. Biosynthesised from squalene via lanosterol.

Common mistakes and connections: Students think steroids are only anabolic drugs. Another error is forgetting that cholesterol is both essential (membranes) and risky. Substituent methyl groups at C18 and C19 are angular. Links to terpenes, lipids, stereochemistry of fused rings and chair conformations.

Visualization of Steroids

The steroid skeleton fuses three six-membered rings and one five-membered ring, the same core in cholesterol and sex hormones. The interactive lesson shows this as a live animation or chart.

Worked example and use case

Testosterone and oestradiol share the same steroid skeleton but differ in the substituents on ring A, giving very different hormonal effects.
