In-depth explanation of Pyridine

Pyridine is a six-membered aromatic heterocycle containing one nitrogen atom that is sp2 hybridized. Its nitrogen lone pair lies in the plane of the ring and is not part of the six-electron pi system, so it is available to accept a proton. Pyridine is therefore a weak base, with the conjugate acid having a pKa of about 5.2. The electronegative nitrogen withdraws electron density from the ring, so pyridine is deactivated toward electrophilic substitution, which occurs mainly at the 3 position.

Going deeper: Pyridine

Key ideas: Six-membered aromatic ring with one N; the lone pair is in an sp2 orbital in the plane, so it is available for protonation (pKa of conjugate acid 5.2, weaker than aliphatic amines). Electron-poor ring: electrophilic substitution is hard (needs harsh conditions, goes to C3), nucleophilic substitution happens at C2 and C4 (Chichibabin reaction with NaNH2). Miscible with water and used as base and solvent.

Common mistakes and connections: Students expect pyridine to nitrate as easily as benzene or to substitute at C2 under electrophilic conditions. Another error is saying the nitrogen lone pair contributes to aromaticity; it does not. Links to heterocycles, nucleophilic aromatic substitution, vitamin B3 and alkaloids like nicotine.

Visualization of Pyridine

Pyridine is a weak base (pKa of conjugate acid 5.2) because its lone pair sits in an sp2 orbital outside the aromatic pi system. The interactive lesson shows this as a live animation or chart.

Worked example and use case

Pyridine is used as a solvent and as a precursor of nicotinic acid, a vitamin.
